Publication Type: Abstract Only Publication Acceptance Date: 9/21/1995 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: Our objective is the creation of a genetic linkage map of soybean that includes SSR, RFLP, and RAPD DNA markers as well as classical genetic markers including loci controlling morphological, pigmentation, disease resistance and other traits. Loci controlling chlorophyll deficiency provide a group of easily scorable markers for map construction. Crosses were made between a normal parent and genotypes homozygous for 12 different chlorophyll deficiency alleles as well as 11 additional chlorophyll deficients in the Genetic Type Collection. DNA was extracted from bulked homozygous recessive (chlorophyll deficient) F2 plants. The SSR allele(s) present in the bulks at 32 loci were compared to the allele present in each homozygous parent. In 12 instances the SSR allele from the chlorophyll deficient parent was clearly the predominant allele in the bulk. In these instances of putative genetic linkage, the map distance between the SSR and chlorophyll deficient locus was determined by analyzing the SSR alleles present in individual homozygous recessive plants. |
