Title: IMMUNE RESPONSE OF MICE CHALLENGED WITH BRUCELLA ABORTUS STRAIN 2308 AFTER ORAL VACCINATION WITH ATTENUATED SALMONELLA TYPHIMURIUM EXPRESSING BRUCELLA GROEL

Interpretive Summary:

Technical Abstract: Resistance to infection with pathogenic Brucella abortus strain 2308, lymphocyte proliferative response, and antibody titer to a 60 kDa A. abortus GroEL heat shock protein were evaluated in BALD/c mice orally vaccinated with recombinant Salmonella typhimurium expressing the complete A. abortus GroEL gene product. Control mice were inoculated with saline, B. abortus strain 19, or nonrecombinant S. typhimurium. Mice were evaluated for serologic and blastogenic response to GroEL at 3 and 10 weeks after vaccination and at 1, 3, 9, and 18 weeks post challenge with B. abortus strain 2308. Antibody titers of GroEL vaccinated to GroEL (measured by ELISA) at 3 weeks post vaccination were not significantly different from other vaccination treatments. At 10 weeks post vaccination the GroEL titer was significantly higher (P<.05) in GroEL vaccinates than S19 or saline vaccinates. Antibody titers to GroEL were significantly high (P<.05) at 3 and 18 weeks post challenge in GroEL vaccinates. Blastogenic response to GroEL did not differ by vaccination treatment before challenge. After challenge, S19 vaccinates had a higher blastogenic response to GroEL than did any of the other vaccination treatments (P<.05). Mice vaccinated with GroEL were not as well protected from subsequent infection with B. abortus strain 2308 as B. abortus strain 19 vaccinates.
