Title: PATTERN OF GRANULOSA CELL PROLIFERATION DURING ANTRAL FOLLICULAR GROWTH IN MEISHAN GILTS

Interpretive Summary:

Technical Abstract: Meishan gilts were ovariectomized on Day 15 to 19 of the estrous cycle or 24 to 30 h after observed estrus (post-LH, PLH). Two h before ovariectomy, each gilt received an iv injection of bromodeoxyuridine (BrdU; a thymidine analog). BrdU was immunolocalized in histological section of follicles greather than or equal to 3 mm in diameter, and the labeling index (LI; % of nuclei staining for BrdU) within the basal, middle, and antral thirds o the granulosa was quantified for each follicle. Follicles from Day 19 and PLH gilts had few and no labeled granulosa cells, respectively. Mean follicular diameter (mm) increased linearly (p < .01) from 4.0 at Day 15 to 7.9 at PLH. Antral volume (mm3) increased linearly (p<.01) from 12.9 at Day 15 to 100.5 at Day 19, resulting in 2.8 volume doublings and a doubling time of 1.4 days. Number of granulosa cells/follicle (x 1,000,000) increased linearly (p<.01) from 4.2 at Day 15 to 15.4 at PLH, resulting in 1.5 cell doublings and a doubling time of 3.3 days. Thus, as follicles matured, cell proliferation decreased in the basal and middle but not the antral area of the granulosa. In addition, follicular growth resulted primarily from antral expansion.
