Title: FINE MAPPING OF QTLS CONTROLLING GRAIN YIELD IN MAIZE

Interpretive Summary:

Technical Abstract: We conducted a large study to determine genetic factors important for heterosis in maize (Genetics 132:823-839). QTL analysis of an F3 population derived from the two elite maize inbreds B73 and Mo17 showed the region around Amp3 on chromosome five accounted for approximately 30% of the variation for grain yield. To further characterize this area a set of BC2S1 isolines was developed. Fifty of these lines contained overlapping B73 genomic fragments between Pgm2 and Amp3 in an otherwise Mo17 background. A similar series of lines contained Mo17 overlapping genomic fragments in a B73 background. The genotype of the overlapping segment in each isoline was determined using 16 RFLP and 2 isozyme markers. Total grain yield and yield component traits (ear and kernel characters) were measured on the isolines at three locations. Using single factor ANOVA we have determined that at least three QTLs controlling grain yield exist in this region. In addition, by comparing individual lines in a manner similar to deletion analysis we have mapped one effect to a region between Amp3 and the sequence detected by bnl5.71. Positive grain yield effects in this region are mostly associated with increased ear length.
