Publication Type: Abstract Only Publication Acceptance Date: 1/17/1996 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: Ceftiofur is an extended-spectrum beta-lactamase resistant cephalosporin indicated in the treatment of bovine respiratory disease in lactating and non-lactating cattle. Monoclonal antibodies were prepared against ceftiofur using its hydrolyzed form, desfuroyl ceftiofur, as the hapten. A stable monoclonal antibody, Cef-116, of IfG1 was isolated. A competitive inhibition enzyme-linked immunosorbent assay (cELISA) was developed for the quantitative detection of ceftiofur in animal tissues and fluids using a heterologous assay system to provide the most sensitive ELISA. Detection limits for ceftiofur were 0.3 ppb for this assay. Preliminary studies indicate that only a simple aqueous extraction is necessary to detect ceftiofur in fortified bovine tissues. Seventy-five percent recoveries have been observed with spike levels ranging from 0.1 to 1 ppm. Incurred ceftiofur residues in bovine tissues and fluids have currently been produced and analyzed. |
