Due to their extensive use, it is necessary to monitor for residues in various animal-based foods. Current analytical methods for the detection of sulfonamide antibiotics involve solvent extraction and cleanup followed by chromatography. Immunoassays have been developed for some sulfonamide antibiotics but few have utilized monoclonal antibodies. We have developed a sensitive monoclonal antibody-based immunoassay for sulfaphyidine. An aminobenzoic acid sulfonamide hapten-protein conjugate was synthesized and used to immunize five mice. Only one mouse tested positive for antibodies which could be used to detect relevant sulfonamide drugs. A rare antibody-secreting hybridoma was isolated that was highly sensitive for sulfapyridine (IC50=25 ppb) and showed cross reactivity toward other sulfonamides. Results from molecular modeling studies of the hapten and various sulfonamide drugs suggested that antibody recognition was dependent on N**1 aromatic ring substitution and will be further discussed. |
