Publication Type: Abstract Only Publication Acceptance Date: 6/25/1996 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: A solid-phase extraction (spe)/LC method for analysis of niacin in Infant Formula and Wheat Flour was evaluated as an AOAC Peer-Verified Method. The sample is digested in 2.8N H2SO4 at 121C, pH adjusted, filtered and niacin is extracted using an Aromatic Sulfonic Acid-spe column. Niacin is eluted with 0.25M sodium acetate/acetic acid (pH=5.6) buffer using a specially designed multi-sample vacuum manifold. Determination of the eluted niacin was done by anion-exchange LC. Collaborative study of this method using NIST SRM-1846 Infant Formula (certified for niacin) and NIST RM-8438 Soft Winter Wheat (published values) as test materials is being done as part of the Peer-Verified Method process. Niacin values for these reference materials were 85-108% of label values. AOAC recommends use of reference materials with application of AOAC methodology, when available. The SRM/RM materials utilized as test samples for this Peer Validation study are readily obtainable from NIST. Thus, these materials were chosen as the tes materials for the validation process in order to assure availability of appropriate reference materials in future implementation and use of this Peer Validated Method. Development/testing of appropriate reference materials in conjunction with the method validation process should be emphasized as an important part of future validation studies. |
