Semen used for AI has a great potential for the transmission of infectious diseases. Virus isolation (VI) has been commonly used to diagnose viral diseases of animals. However, detection of virus in boar semen using VI is not sensitive because of cytotoxic effect of semen on cells and low amount of viral particles in semen. Thus, we developed a highly sensitive detection method using nested polymerase chain reaction (PCR) for pseudorabies virus (PRV), porcine reproductive and respiratory syndrome virus ()PRRSV), and porcine respiratory coronavirus (PRCV). We report the detection of PRV DNA and PRCV RNA in semen from experimentally infected boars, and multiplex PCR using one-step RT-PCR coupled with nested PCR for the simultaneous detection of PRV, PRRSV, and PRCV (m)RNAs in boar semen. |
