To obtain saliva proteins, about 200 adult potato leafhoppers were fed on diet cubes comprised of agarose and sucrose. Proteins were electro-eluted from the diet cubes into a buffer. To obtain salivary gland proteins, glands were dissected from adult potato leafhopper, and the soluble protein content wa actively diffused into a buffer. The protein content from both saliva and salivary glands were separated by charge and size using vertical and continous flow electrophoresis. |
