Interpretive Summary:

Technical Abstract: The nucleotide sequence of a BamHI-F fragment in the short unique (Us) region of MDV serotype 2 (strain HPRS24) was determined. Seven potential open reading frames (ORFs) were identified. Six of then had counterparts in the Us region of herpes simplex virus type 1, including US2, US10, protein kinase (US3), and glycoprotein gD, gI and gE. The other ORF was homologous to MDV-specific SORF3 which has been reported in other MDV serotypes. To investigate the antigenic property of MDV2 homologs of gI and gE, the potential coding regions were expressed by a baculovirus expression system. The gI homolog was immunoprecipitated as two specific 45 and 43 kDa glycoproteins and an unglycosylated 35 kDa precursor polypeptide by anti-HPRS24 chicken serum. The gE homolog was expressed and glycoproteins with multiple sizes from 72 to 66 kDa and an unglycosylated 45 kDa precursor polypeptide. Further, the expressed gI and dgE homologs reacted with all chicken-antisera raised to each of the three serotypes of MDV, suggesting that these glycoproteins are actually expressed in infected chickens by all three serotypes of MDV and conserve common antigenic epitope(s) beyond serotypes.
