Interpretive Summary:

Technical Abstract: Glutamine synthetase (GS) catalyzes the assimilation of ammonia into glutamine and is a key enzyme in nitrogen metabolism. The pedicel region of the maize kernel contains a kinetically and physically unique GS isoform whose activity increases during kernel development. Molecular cloning of GS1 genes expressed in the kernel pedicel and analysis of their expression pattern established that the maize GS1-2 gene likely encodes this kernel pedicel-specific GS isoform. To further study the pattern and regulation of GS1-2 expression in the pedicels, genomic clones for GS1-2 were isolated from a maize genomic library. A genomic clone containing the entire GS1-2 gene, including the 5' flanking sequences, was characterized. The upstream promoter region was sequenced and the transcription start site mapped using primer extension and 5' RACE. Results of analysis of the GS1-2 gene promoter using the kernel transient expression system will also be presented.
