Interpretive Summary:

Technical Abstract: Soybean Cyst Nematode (SCN) resistance in soybean is a complex oliogenic trait. One of the most important resistance genes, rhg1, has been mapped to a distal region of molecular linkage group G. In order to develop technology for marker assisted selection and for map-based cloning of the gene, we have developed a high resolution map of the genomic regions adjacent to rhg1. Four different segregating populations have been used to identify informative recombination events for mapping markers near rhg1. Bulk segregant analysis has placed six AFLP in ca. 1 cM proximal to the gene. In addition, a fortuitous microsatellite marker is located 0.5 cM distal to the gene. Physical mapping of the region has been partially completed through the construction of BAC contigs anchored upon the markers most closely linked to rhg1. The contigs have provided additional molecular probes that detect RFLPs within 0.2 cM of the gene. Large populations that segregate for the gene have been screened to provide additional recombinants to map markers accurately to within 0.05 cM. The close proximity of the markers and the advancing BAC contig provides the resources for the ultimate cloning of rhg1.
