Interpretive Summary: This study was performed to determine if intact sperm storage tubules could be successfully isolated from the uterovaginal junction mucosa of the turkey. Large White turkey hens were inseminated and sacrificed 24-48 hr later. Oviducts were isolated and removed, and the uterovaginal junction mucosa was dissected and enzymatically digested. Morphologically normal, intact sperm storage tubules that contained slowly oscillating sperm were successfully recovered. This method for the isolation of intact sperm storage tubules can be readily applied for in vitro culture studies as well as for the extraction of DNA and RNA from the SST epithelium.
