Research Project: Engineering Sustainable 3D Scaffolds for Enhancing Delivery and Growth of Probiotics in the Gut

Location: Healthy Processed Foods Research

Project Number: 2030-41440-008-013-R
Project Type: Reimbursable Cooperative Agreement

Start Date: Jun 1, 2023
End Date: May 31, 2025

Objective:
1: Assess enhancement in survivability of probiotics in simulated gastro-intestinal environment of diverse probiotic bacteria using 3d delivery scaffolds, a simulated fecal transplant, of different composition derived from fruit and pulses. 2: Assess role 3D delivery scaffolds and in-situ growth of bacteria in these scaffolds for persistence and muco-adhesion of probiotic bacteria in the gut. 3: Assess efficacy of optimized probiotic delivery formulations in reducing metabolic syndrome associated with obesity and high fat diets.

Approach:
Measure in-vitro mucoadhesion of plant cell ghosts containing beneficial microbes after simulated GI digestion followed by in-vivo evaluation of persistence compared to controls in mice. Intestinal, cecal and colonic contents and organs will be collected after oral administration of the 3D scaffold probiotics. Microscopic analysis of plant carrier and bacteria will be conducted. Physiological response to treatment will be compared to controls for effects on metabolic dysfunction.
