Objective:
Quantify the presence of: 1) AMR gene isolates (ermB, sulI, intlI, and blactx-m-32), and 2) enteroviruses (EV), rotavirus (RV) and adenovirus (AdV) genes as water quality indicators in leachate of karst and non-karst soils in successive rainfall simulations from intact large soil columns.
Approach:
Cooperator will quantify AMR isolate presence in collected leachate via polymerase chain reaction (PCR) following poultry litter application and three successive simulated rain events. Virus gene detection will occur by cooperators using quantitative-PCR methods. These data will be scaled to identify AMR isolate leaching data from intact soil columns with an ARS-developed digital mapping process based on topography to develop precision management practices by linking to regional digital soil maps for identifying karst topography presence and soil profile AMR movement data.
