Objective:
To assess the effect of knocking out GDF9 and/or BMP15 on ovarian function and follicular progression in Japanese quail.

Approach:
The Cooperator will confirm the sequences of GDF9 and BMP15 in Japanese quail and will construct adenoviral vectors with guide RNAs targeting these genes. The process will be tested in cell cultures for editing efficiency by the Cooperator. After vectors with efficient guide RNAs are selected, the adenovirus will be produced commercially. The adenovirus will be injected by the PI into Japanese quail blastoderms to produce gene-edited chimeras. These chimeras will be bred to obtain heterozygous quail that will further be mated to obtain GDF9 and BMP15 knockout females. Follicular development will be analyzed after the birds reach sexual maturity.
