Research Project: SSBRI Molecular Evolution of Beet Necrotic Yellow Vein Virus and Rhizomania Disease Epidemiology Compromising the Host Resistance in Sugar Beet

Location: Sugarbeet Research

Project Number: 3060-21000-045-018-S
Project Type: Non-Assistance Cooperative Agreement

Start Date: Sep 1, 2023
End Date: Dec 30, 2024

Objective:
Evaluate field soil samples for rhizomania and recover viruses that compromise rhizomania. 2)-Identify the viral variants of Beet necrotic yellow vein virus that compromise the host resistance. 3)-Determine the ability of the identified BNYVV variants in compromising host resistance provided by Rz1, Rz2, and Rz1 plus Rz2 of sugar beet varieties.

Approach:
The cooperator will carry out the following: 1)-Application of high-throughput sequencing (HTS) on the root samples obtained from soil-bait sugar beet plants using different sugar beet genotypes and that includes RNA extraction, RNA-Seq library construction, and HTS using Illumina platform. 2)- Bioinformatic analyses of raw data obtained from HTS to identify the variations on the BNYVV genome (RNA1, RNA2, RNA3, and RNA4) that can compromise the host-resistance. 3)-Reverse genetics approaches to determine the ability of the identified resistance-breaking variants in compromising the host resistance provided by Rz1, Rz2, and Rz1 plus Rz2 of sugar beet varieties.
