Objective:
The objective is to conduct a quantitative analysis of carotenoids in foods to be reposited in the USDA FoodData Central (FDC) by: 1. Developing and validating a quantitation method for carotenoid extraction and separation via high performance liquid chromatography (HPLC) for use across different types of foods. 2. Applying this method across fruits and vegetables acquired for inclusion in the FDC. 3. Making available to investigators and the public a detailed protocol for the method and its validation to facilitate harmonized analytical approaches for phytonutrients in foods.

Approach:
In collaboration with ARS scientists at the Beltsville Human Nutrition Research Center (BHNRC), Children's Nutrition Research Center (CNRC) at Baylor College of Medicine, and Arkansas Children's Nutrition Center (ACNC) at U Arkansas and scientists at the Virginia Polytechnic Institute & State University, OSU scientists will develop methods for sample handling, carotenoid extraction and quantitative analysis using HPLC with validation across laboratories. Foods of interest will be acquired, processed and analyzed for inclusion in the USDA FoodData Central. Research on optimizing procedures for different food matrices and on developing high-throughput extraction and analyses will facilitate the timely and accurate acquisition of data.
