Objective:
1. Discover and characterize the role of plant phenolics in improving resistance of diverse probiotic bacteria to gastric and intestinal barriers (this may be characterized based on measuring changes in gene expression and metabolism of the probiotic bacteria). 2. Demonstrate in-vivo efficacy of the optimal combinations of plant phenolics and probiotics in influencing delivery and persistence of probiotic bacteria in-vivo. 3. Determine the influence of synergistic combination of plant phenolics and probiotics in the gut in influencing gut health based on improved probiotic function and metabolism of plant phenolics (this may be determined based on gut microbiome, tissue inflammation status and if possible metabolite analysis of probiotic bacteria).

Approach:
Experimental Approach: This aim will be subdivided into two sub-aims (1) includes short term in-vivo experiments to evaluate the survivability, persistence and adhesion of orally gavaged probiotic cells with and without infused plant extracts and (2) Long-term in-vivo experiments to evaluate the influence of synergistic combination of plant phenolics and probiotics in improving high fat diet induced metabolic disorders and gut microbiome.
