Interpretive Summary:

Technical Abstract: The objective of this study was to determine the functional capacity of individual amino acids (AA) as regulators of protein degradation (PD) in rainbow trout primary myoblasts. Incubation of cells in minimal essential media (MEM) lacking essential AAs (EAA) increased protein degradation by 1.5-fold compared to MEM containing a complete AA profile. Addition of excess leucine or phenylalanine attenuated this response by approximately 50%. However, PD increased when lysine or valine were supplied in a 10-fold excess relative to other EAA, suggesting that a lysine or valine imbalance has negative effects on muscle protein retention. Protein degradation increased in leucine deficient media and was reduced in a dose dependent manner with the addition of leucine from 10 nM – 1 mM, indicating that leucine is rate-limiting for PD. Collectively these findings support that individual AAs directly regulate PD in muscle, albeit in an AA-specific manner.
