Thus, an OB protein presumably has a central role in regulating body fat. We have used a cDNA clone of the rat ob gene to map on bovine homologue to chromosome 4 using RFLP analysis. Digestion on bovine genomic DNA with Bgl II resulted in polymorphic bands at 8.2 and 10.5 kb. Fifty percent of reference family parents were heterozygous for these bands. This is a first step in our efforts to characterize the role of OB protein in controlling appetite and fat deposition in livestock. |
