Publication Type: Abstract Only Publication Acceptance Date: 1/29/1996 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: There are numerous instances in which markers based on tri- or tetranucleotide repeats would be very useful in cattle because they are generally much easier to genotype than markers based on dinucleotide repeats. However, unlike humans, the bovine genome contains very few tri- or tetranucleotide microsatellites that are suitable for marker development. A small-insert bovine phagemid library was enriched for clones containing CCA repeats. Positive clones (300) from a primary screening of colony lifts were picked into 96 well plates which also contained clones with known repeat lengths: (CCA)6, (CCA)7, (CCA)9. Upon rescreening, 60 clones that gave strong hybridization signals compared to the (CCA)6 control were sequenced and 40 were confirmed to have (CCA) >6. Sixteen were suitable for primer design and 5 (1.6% of 300 positive clones) yielded informative results. Three of these are sufficiently polymorphic to be useful in providing reliable genotype information in situations wher DNA from families is lacking. |
