Publication Type: Abstract Only Publication Acceptance Date: 7/1/1996 Publication Date: N/A Citation: N/A Interpretive Summary: Technical Abstract: A bovine chromosome 11 (BTA11) painting probe and a phage library was developed by chromosome microisolation and microcloning of BTA11 utilizing a bovine cell line carrying a chromosome translocation t(X;11)(unpublished data). The library was screened for clones containing microsatellite sequences using end labeled (AC)12 oligos as probes. Sequencing was done in an ABI automated sequencer. Primer pairs for 17 microsatellite clones containing 14 or more perfect AC repeats have been developed. These microsatellites were used to generate a BTA11 genetic map based on the MARC reference population. Four bovine YAC clones, each containing a genetically mapped microsatellite were physically assigned to chromosome 11 by fluorescent in situ hybridization. This resulted in an anchored, oriented and integrated physical and genetic map for this chromosome. |
