Interpretive Summary:

Technical Abstract: C-X-C chemokines are inducible proteins expressed during the early phases of infection and are chemotactic to polymorphonuclear leukocytes. The purpose of this study was threefold: 1) define SNPs for bovine chemokine genes, 2) characterize their allele frequencies in reference populations, and 3) localize their position on the bovine genetic map. SNPs were identified by first designing oligonucleotide primers to amplify introns and untranslated regions of chemokine genes. PCR amplification products from 24 MARC bovine reference parents were sequenced and analyzed for nucleotide polymorphisms. Candidate polymorphisms were verified by characterizing allele frequencies in the progeny of the reference parents. Linkage analysis showed clustering of several C-X-C chemokine genes on bovine chromosome 6. A similar gene cluster is present on human chromosome 4, suggesting conservation of synteny for this region of the respective genomes. Genetic variation in bovine chemokines may be useful for identifying alleles associated with infectious disease traits in livestock animals.
